Characterization of crude acidic xylanase from Aspergillus niger

【Abstract】

Xylanase is one of the most important hemicellulases in industry.Commercially available xylanases are usually produced by filamentous fungi such as Aspergillus niger or Trichoderma reesei.Acidic xylanase has played an important role in biofuel production,while using the raw material such as sugarcane bagasses.In this study,characterization of crude acidic xylanase from Aspergillus niger CICC40616 was performed.The optimum temperature ofβ-1,4-endo-xylanase orβ-xylosidase extracted from it was 50-65℃and 75℃respectively.Original activity ofβ-1,4-endo-xylanase was 58.56%at 60℃with an incubation time of 5min,whileβ-xylosidase was 69.08%at 75℃with an incubation time of 15 min.The optimum pH ofβ-1,4-endo-xylanase orβ-xylosidase was both 4.5-5.0.Original activity ofβ-1,4-endo-xylanase was 44.21%at pH 3.5,whileβ-xylosidase was 53.54%at pH 2.1.β-1,4-endo-xylanase was activated by Ca~(2+),Zn~(2+),Sr~(2+),Co~(2+),Mg~(2+),Cu~(2+),EDTA,especially Ni~(2+),but strongly inhibited by Mn~(2+).
